Do I need to manually turn on the microphone prior to sleep or will the sleep report app automatically turn it on? I usually have it turned off during the day to conserve the battery.

By "sleep report" do you mean "Snore Detection"? If so, be forewarned, That is a big battery drain. Unless you are willing to charge your device every day or 2, it is probably best to use just for a day or 2 to get a baseline, then turn off. Then try again in at least a few weeks or when you suspect there might be a change.
In answer to your question, not sure but I think snore detect handles microphone itself.

Follow up: I manually turned off the microphone, and Snore Report was not generated; indicating to me that having the report function turned on would not automatically turn on the microphone once sleep was detected. I will need to turn on the microphone when I go to bed and turn off in the morning to conserve the battery throughout the day.
